WLN-BM.W.34.1Determination of the amount of Escherichia coli by means of MPNin accordance with NEN-EN-ISO 9308-3
Principle:A diluted sample is pipetted in a microtiter plate, containing dehydrated medium with 4-Methylumbelliferyl-β-D-glucuronide (MUG). The microtiter plate is examined in the dark under a UV lamp at a wavelength of 365 nm, after an incubationof 36 hours at 44 °C. The presence of E. coli is indicated by a blue fluorescence as result ofthe hydrolysis of MUG. The results are expressed in MPN (Most Probable Number) per 100 ml.
Determination:E.coli using the MPN method
Performance characteristics:
Matrices qualified by the Dutch RvA:Surface water
Reporting limit:15 kve/100ml, matrix dependent
Measurement uncertainty:60%
Sustainability:1 day , if conserved with NTA and Na-thiosulphate
Sampling bottle:Plastic , white , 500 ml , conservation with NTA and Na-thiosulphate (fill up to the neck of the bottle)
