WLN-BP.W.08.2Determination of the presence of Escherichia coli; RT-PCR (Reverse Transcriptase PCR)in-house method
Principle:The sample is filtered through a suitable membrane filter. Next the bacteria material present on the filter is isolated, purified and made fit for RT-PCR analysis. RT-PCR stands for Reverse Transcriptase - Polymerase Chain Reaction. It is a primer-dependent technique in which, the present RNA is converted into copyDNA, which next will serve as target for the subsequent PCR amplification. The labelling of the synthetically obtained cDNA makes it possible to demonstrate thepresence of the bacterium.
