WLN-BM.W.33.1Determination of the number of enterococcus by means of MPNin accordance with NEN-EN-ISO 7899-1
Principle:A diluted sample is pipeted in a series of microtiter plate cups, containing dehydrated medium that contains 4-methylumbelliferyl-ß-D-glucoside (MUD). The microtiter plate is examined in the dark under aUV lamp at a wavelength of 365 nm, after an incubation of 36 hours at 44 °C. The presence of Enterococcus is indicated by a blue fluorescence as result of the hydrolysis of MUD. Theresults are expressed in MPN (Most Probable Number) per 100 ml.
Determination:Enterococcus using MPN method
Performance characteristics:
Matrices qualified by the Dutch RvA:Surface water
Reporting limit:15 kve/100ml, matrix dependent
Measurement uncertainty:60%
Sustainability:1 day , if conserved with NTA and Na-thiosulphate
Sampling bottle:Plastic , white , 500 ml , conservation with NTA and Na-thiosulphate (fill up to the neck of the bottle)
