WLN-BP.W.06.2Determination of Legionella pneumophila by means of DNA methodin accordance with NEN 6254+C1
Principle:The sample is filtrated through a suitable membrane filter. The bacteriological material on the filter is isolated, purified and made suitable for PCR analysis. A known amount of internal standard is added to the mixture. The result of this reprocessing is corrected for the yield of the internal standard. The amount of the proved L.pneumophila is expressed in numbers of copies DNA per liter. The limit of quantification of this method is 100 cDNA/l but depends on the filtration volume and reprocessing efficiency.
Determination:Legionella Pneumophila (DNA-method)
Performance characteristics:
Matrices qualified by the Dutch RvA:Drinking water, Cooling water, Swimmingpool water
Reporting limit:100 cDNA/l, matrix dependent
Measurement uncertainty:75%
Sustainability:1 day , if conserved with NTA and Na-thiosulphate
Sampling bottle:Plastic , white , 375 ml , conservation with NTA and Na-thiosulphate (half warm and half cold water)
