WLN-BM.W.06.2Determination of Legionella by means of membrane filtrationin accordance with NEN-EN-ISO 11731
Principle:The sample is filtered through a suitable membrane filter. Next the filter is treated ultrasonic. For elimination of disturbing flora one part of the concentrate is pasturised at 50°C and one part istreated with acid. The non pasturised, pasturised and acid treated concentrate is placed and spreaded on different solid, selective mediawith selective characteristics and incubated at 37 °C for 7-10days.The Legionella bacteria will form characteristic colonies. The suspectcolonies are confirmed by MALDI-TOF.
Determination:Legionella (poorly filterable)
Performance characteristics:
Matrices qualified by the Dutch RvA:Cooling water, Surface water
Reporting limit:100 kve/l, matrix dependent
Measurement uncertainty:60%
Sustainability:2 days , if conserved with NTA and Na-thiosulphate
Sampling bottle:Plastic , white , 375 ml , conservation with NTA and Na-thiosulphate (half warm and half cold water)
